Organoid development and tissue regeneration are regulated by both intrinsic cellular properties and the surrounding biological microenvironment, or bio-niche. The composition and architecture of the extracellular matrix (ECM), biochemical and biophysical cues, and cell–cell interactions play essential roles in directing stem cell fate, tissue formation, and functional maturation. Understanding and engineering tissue-specific microenvironments are therefore central to developing functional organoids and regenerative platforms.

This seminar will introduce bioengineering strategies for creating supportive niches for organoid development and tissue regeneration, with a particular focus on ECM-based microenvironmental design. It will explore how tissue-derived ECM, three-dimensional architecture, and the controlled presentation of biochemical and biophysical signals can guide cellular differentiation and maturation. Strategies for modulating cell–matrix and cell–cell interactions to recapitulate native tissue physiology and establish environments conducive to regeneration will also be discussed.

The seminar will further examine the opportunities and remaining challenges in applying bio-niche engineering to organoid-based disease modeling, drug testing, tissue regeneration, and cell therapy.
